Metabolic Study in GDF11-Treated Young and Old Male C57BL/6 Mice Fed High-fat

Description Aging is associated with declined metabolic function. Interventions which
improve metabolic function in aged individuals have great therapeutic potential.
We would like to determine whether treatment to restore youthful circulating
levels of a protein that has been shown to decline with age can have durable
improvements in metabolic function in both young and old mice. These studies
will demonstrate the therapeutic potential of this treatment in aged
individuals.

This study is approved for the Metabolism Core of UMass MMPC and will involve 50
HFD-fed C57BL/6J mice for drug treatment (GDF-11 = vehicle), body composition,
and hyperinsulinemic-euglycemic clamp.

Male C57BL/6J mice from JAX will be ordered by Dr. Lee and sent to the UMass
MMPC. There will be 20 mice at 8 weeks of age and 30 mice at 24 months of age at
the time of arrival at UMass MMPC. Following arrival, mice will receive a HFD
and a daily treatment (ip injection) with saline or GDF-11 for 7 days prior to
the clamp experiment.

This study will be led by Dr. Hye-Lim Noh and Dr. Taekyoon Kim.
