Hyperinsulinemic Euglycemic clamp order # 20704 2015

Description Hyperinsulinemic-euglycemic clamp: Insulin is continuously infused at a constant
rate during 155min (see data for infusion rate), and glucose is infused at a
variable rate to maintain euglycemia. 3H-glucose is infused to assess glucose
fluxes. At 120min, a bolus of 14C-2-deoxyglucose is given to assess
tissue-specific glucose uptake. Mice are sacrificed at 155min.

Mouse were fasted for 5h prior to experiment.

Groups:
Controls are wildtype littermates
WT and KO animals were subjected to clamps after having been treated with saline
or an FGF21 agonist (bispecific anti-FGFR1/KLB Ab) which was injected 3 days
prior to a study (EBioMedicine2 (2015)730–743)
Male WT and KO

Purpose of the experiment: This experiment will determine whether the groups of
mice have different insulin sensitivity, and will determine sites of insulin
resistance.
