Experiment

Circulating beta-hydroxybutarate and adiponectin concentrations in Rdh10 KO mice
Summary Data Summary
Investigator Napoli, Joseph
Description Rdh10KO mice have decrease in atRA in liver and adipose tissue, and exhibit a
severe metabolic phenotype when fed high fat diet with 4IU vitamin A/g. Mice
develop increased adiposity in the bone marrow and liver steathosis. This is a
valuable mouse model to study osteoporosis.

Rdh10 KO heterozygote mice develop severe metabolic syndrome when fed AIN-93G
diet with 50% of calories derived from fat (HFD). Both males and females develop
increased adiposity compared to WT littermates, and males develop liver
steathosis by 16 weeks of age. Female mice show bone loss at 16 weeks of age on
either low fat or high fat diet. Curent study was designed to measure serum
adiponectin levels after 16 h fasting period. Mice had ad lib access to HFD and
water. Mice were put on the HFD at 4 weeks of age. Blood samples were collected
via tail tip after 16h fast, and serum was separated from colagulated blood by
centrifugation. Mice were anesthetized with 2% isofluorane mixed with oxygen.
