Description Apoe knockout: Insertion of a neomycin resistance cassette
deleted part of exon 3 and part of intron 3 of the Apoe
gene. Plasma from homozygous mutant mice gave no detectable
immunoprecipitate by the Ouchterlony double immunodiffusion
test using a rabbit antibody to rat APOE.

ROSA26: The targeting vector was designed with a CMV
enhancer/chicken beta-actin core promoter (pCA) driving
expression of a loxP-flanked, N-terminal membrane-tagged,
optimized DsRed fluorescent protein variant (called
tandem-dimer-Tomato or tdTomato) sequence followed by a
polyadenylation signal. Immediately distal to the second
loxP site is an N-terminal membrane-tagged, enhanced green
fluorescent protein (EGFP) sequence itself followed by a
polyadenylation signal. An frt-flanked neo cassette was also
located distal to the expression vector. Red fluorescence is
detected in all tissues tested. When mice are bred to Cre
expressing mice, the floxed region is excised in
Cre-expressing tissue and this allows expression of the EGFP
cassette.

Tgfbr1 knockin: Exon 3 was left flanked by single loxP sites
after the removal of an upstream floxed neo cassette by the
transient expression of cre recombinase in ES cells.

Tgfbr2 knockin: This allele is derived from Tgfbr2tm1Hlm.
Expression of cre recombinase at the one cell stage resulted
in the excision of the floxed region containing the neo
transgene. Two single loxP sites were left flanking exon 2.

Cdh5-cre: The tamoxifen-inducible cre/ERT2 sequence was
inserted into a PAC (P1 artificial chromosome) containing
the VE-cadherin (Cdh5) gene to generate the transgene
construct.
