Common Name GHRH-Cre ERalpha floxed
Description GhrhCre knock-in mice have a viral P2A oligopeptide fused to
a Cre recombinase gene replacing the stop codon of the
growth hormone releasing hormone locus (Ghrh). GHRH is
expressed in hypothalamic neurons. These neurons are
involved in control of food intake, glucose homeostasis, and
growth. GhrhCre knock-in mice have Cre recombinase expressed
in areas of the hypothalamus, including the arcuate nucleus,
with minimal expression in a few regions in the midbrain and
hindbrain. Cre replaces the stop codon, and the presence of
a P2A cassette allows expression of both GHRH and Cre.
Homozygous mice are viable and fertile. When these mice are
bred with mice containing loxP-flanked sequence,
cre-mediated recombination will result in deletion of the
floxed sequences in the cre-expressing cells of the
offspring.
The Kiss1 (KiSS-1 metastasis-suppressor) gene encodes a
metastasis suppressor protein that is active in melanomas
and breast carcinomas, and also acts to promote secretion of
gonadotropin-released hormone. These transgenic mice express
Cre recombinase, under the control of the mouse Kiss1
promoter. When crossed with a strain containing loxP site
flanked sequence of interest, Cre-mediated recombination
results in tissue-specific deletion of the target.
Recombination in founder line J2-4 animals occurs in Kiss1
expressing neurons of the arcuate nucleus and anteroventral
periventricular nucleus of the hypothalamus as well as in
the placenta, pancreatic beta-cells and Leydig cells. The
Cre recombinase activity pattern mimics the endogenous gene
expression (mRNA) pattern. The Donating Investigator reports
homozygous mice are viable and fertile.
The mutation is a derivative of Esr1tm1Mma. The TK-neo
cassette was deleted by Cre mediated recombination. In the
resulting mice, exon 3 is flanked by loxP sites.
For deletion of ERa or AR signaling in GHRH-expressing
neurons, the GHRHCre mouse was crossed with a mouse carrying
the loxP-modified Esr1 (ERafl/fl) or Ar (ARfl/fl) alleles.
