Effect effects of dietary starch and probiotics on plasma LBP in DIO mouse model

Description C57BL/6J mice at 6 weeks old were obtained from the Jackson Laboratory (The
Jackson Laboratory, Sacramento, CA, USA). Mice were singly housed in cages lined
with a 4:1 ratio of TEK-Fresh Laboratory Animal Bedding (Harlan, Madison, WI)
and paper chip (Shepherd specialty papers, Watertown, TN). Animals were
maintained on a 12 h light/dark cycle (lights on at 06:00 am) and given free
access to food and water for the duration of the study. For acclimatization, the
mice were maintained for two weeks on a standard mouse chow (Harlan
Laboratories, Inc., Madison, WI). The mice were then fed a custom high-fat (HF)
diet (Harlan, Madison, WI) formulated to contain 45% kcal from animal fat (lard)
for the duration of 9 weeks. The mice were divided into two groups and were
given either 2 × 10^9 CFU of L. plantarum WCFS1 (NCMB 8826)(LP) in 200 µl PBS,
or 200 µl PBS (HFD control), via oral gavage four times per week, for the
duration of 3 months. Groups tested: HF: high fat diet; RS: high fat-resistant
starch diet; LP: high fat diet & gavaged with Lactobacillus plantarum NCMB 8826
every other day at approximately 1X10^9 colony forming units; MU: high fat diet
& gavaged with LP NCMB 8826 that has been genetically modified to not express
three genes (plantaricin E, plantaricin F, and plantaricin I) at approximately
1X10^9 CFUs.
