Experiment

NMR of DIO C57BL/6 and Control Mice
Summary Data Summary
Investigator Jones, Graham
Description These mice are part of a study aimed at evaluating the impact of body
composition on motivated feeding, specifically the lasting effects of having a
history of obesity and then subsequently losing weight. These mice are all
genetically wild type C57BL6 animals, and represent the diet-induced obesity
(DIO) leg of the studies. There are 3 groups of animals with both males (M) and
females (F) comprising each group. The control animals (DIOCM and DIOCF) have
been maintained on normal chow for the entirety of their lives. The DIO (DIOM
and DIOF) and DIO-weight loss (DIOWLM and DIOWLF) animals were weaned onto a
high-fat (36%) high-sucrose (30%) (HFHS) chow to stimulate weight gain,
predominately through the accrual of adipose tissue. After reaching a body
weight criteria (35 g for F, and 45 g for M) the DIOWL group was switched to ad
libitum normal chow to induce weight loss, additional calorie restriction was
used to return body weights down to the level of the DIOC mice; the DIO animals
stayed on the HFHS chow. The mice were then assayed in operant paradigms to
assess motivated feeding. The NMR was conducted following all behavioral testing
and after the animals had been housed with ad libitum access to their respective
diets (normal chow for DIOC and DIOWL groups, and HFHS chow for DIO groups) for
at least two weeks.
