Sable Energy Balance #24532 3/16/18

Description We previously showed that the GLP1 receptor agonist, Exendin-4 (Ex4), reduces
body weight by acute reductions in food intake and chronic increases in energy
expenditure in obese mice. The mechanisms by which Ex4 increases energy
expenditure is not currently known. While weight loss by GLP1 receptor agonists
can be mediated by the brain, there is evidence that Ex4 can increase oxygen
consumption in skeletal muscle tissue directly. However, it is controversial
whether a canonical GLP1 receptor is expressed by skeletal muscle. Further, some
data suggest the possibility of another receptor in skeletal muscle through
which Ex4 can stimulate signaling. In this study, we will be using a global GLP1
receptor KO mouse to determine whether the canonical GLP1 receptor is necessary
for this Ex4-mediated increase in energy expenditure.

Energy expenditure measured by indirect calorimetry in the Promethion by Sable
Systems. Activity, food intake and water intake are also continuously measured.
Measurements were taken for 11 consecutive days.
Running wheels were not present in the cage.
Mice were fed ad libitum.
Water: regular tap water, ad libitum.

Groups:
Male WT (C57BL/6J) mice.
Mice were fed a chow diet (LabDiet PicoLab Rodent Diet 5LOD).
Mice were treated with either PBS or Exendin-4.
