Influence of Myo K64A mutation on metabolic phenotype and energy balance

Description Oxygenated myoglobin has been shown to bind fatty acids or fatty acylcarnitines,
where the Lysine at position 64 is critical to enable binding of fatty acids and
fatty acylcarnitines. However, it is unknown if myoglobin is involved in
whole-body fatty acid oxidation. The objective of this study is to determine if
knocking out myoglobin or inducing a K64A point mutation to prevent lipid
binding influences substrate oxidation (respiratory exchange ratio) and other
metabolic phenotypes in high-fat fed, cold-challenged mice.

Male & female MyoK64A and litter mate controls will have their baseline energy
expenditure assessed by CLAMS at 5 weeks of age. Animals will be placed on
high-fat (HFD) diet at 6 weeks After 5 weeks on diet (age= 11 w) and 12 weeks on
diet (age=17 w) energy expenditure will be reassessed. Energy expenditure will
be assesses at 10 and 22°C followed by body composition analysis by DEXA.
