Hyperinsulinemic Clamp Order #13167 November 2014

Description Hyperinsulinemic-euglycemic clamp: Insulin is continuously infused at a constant
rate during 155min (see data for infusion rate), and glucose is infused at a
variable rate to maintain euglycemia. 3H-glucose is infused to assess glucose
fluxes. At 120min, a bolus of 14C-2-deoxyglucose is given to assess
tissue-specific glucose uptake. Mice are sacrificed at 155min.

Mouse were fasted for 5h prior to experiment.
Water: regular tap water, ad libitum.

Groups:

Purpose of the experiment: This experiment will determine whether the groups of
mice have different insulin sensitivity, and will determine sites of insulin
resistance.

Applicable Research Areas: Metabolism, Insulin resistance.

Male C57BL/6J mice, when fed a high-fat diet, rapidly develop obesity and
manifest increased insulin resistance, hyperglycemia, dyslipidemia, and
hypertension. These are all characteristics of a human with metabolic syndrome.
