Description Insulin resistance is a hallmark of type 2 diabetes and the
mechanism is unclear. Binding to the insulin receptor on the
cell membrane, insulin can activate an intracellular protein
kinase cascade, such as protein kinase B that can
phosphorylate its substrate protein Foxo1 at a serine
residue in the 253rd amino acid, then mediating the effect
of insulin on gene expression that lowers blood glucose
levels and regulates many other metabolic and cardiovascular
functions. To assess the role of Foxo1-Serine 253
phosphorylation in the action of insulin, we generated
Foxo1-S253A knock-in mice, in which the Foxo1-Serine 253 was
mutated to alanine, an amino acids that blocks
phosphorylation from the protein kinase B in insulin action.
