Description MC4R full body KO using an inserted transcriptional blocker
with selective neuronal reactivation using ChAT-Cre

Mc4r mutation: A loxP flanked PGK-neomycin selection marker
inserted in the 5' UTR created a null allele. Cre
recombinase deletes the selection marker, restoring normal
activity of the allele. Mice fail to respond to MC4R agonist
MTII. This is a null allele that can be "reactivated" by cre
recombinase.

Chat-cre mutation: An optimized internal ribosome entry
sequence (IRES) fused to cre recombinase followed by a
neomycin resistance cassette flanked FRT sites was inserted
after the ChAT stop codon in BAC (bMQ185k21) using
homologous recombination in bacteria. A genomic ChAT
fragment with 4kb homology arms on either side of the
IRES-Cre insertion site was lifted out from the BAC using
reverse recombineering techniques and transformed into ES
cells. The neo cassette was excised with Flp recombinase.
Cre recombinase activity is reported in all cholinergic
neurons. This allele generated from Chattm2(cre)Lowl.
