Circulating insulin and lipid levels in Lepr KO mice treated with niclosamide
derivatives

Description BKS.Cg-Dock7m +/+ Leprdb/J mice received daily oral gavage of vehicle or
1.0mg/kg or 2.5mg/kg body weight of WX53 or 2.5mg/kg or 5.0mg/kg body weight of
Y03 for 3 weeks. C57BL6/J mice with Research Diets-D12492 received daily oral
gavage of 1.0mg/kg or 2.5mg/kg or 5.0mg/kg body weight of WX53 or 2.5mg/kg or
5.0mg/kg body weight of Y03 for 3 weeks. 5 hour-fasted blood was collected
through tail vein bleeding at the end of the treatment. Plasma was obtained by
centrifugation at 1000g for 10-15min at 4? . Whole blood and plasma stored at
-80? for further analysis. WX53 and Y03 are from a group of mitochondrial
uncouplers that can stimulates oxygen consumption. Hypothesis: oral
administration of mitochondrial uncouplers could reduce blood glucose level and
improve insulin resistance. The objective of the study is to investigate the
effect of mitochondrial uncouplers on the treatment of type 2 diabetes.
