Experiment

Effect of Dinitrophenol (DNP) and mitochondrial uncouplers on Energy
expenditure.
Summary Data Summary
Investigator Sinick, Jonah
Description The objective of this study is to determine if a single 5, 10 or 20mg/kg IP dose
of 2-dinitrophenol (DNP) or mitochondrial uncoupler (BAM15 or Niclosamide) are
sufficient to elicit a change in murine energy expenditure. Male c57BL6/J ~9
weeks of age are acclimate for 1-2 week on std. vivarium chow (Teklad 2918)
before undergoing CLAMS. After acclimation to CLAMS, mice are injected with
saline on day 1 and compound of interest on day 2 of energy expenditure
analysis. Upon completion of CLAMS, body composition is assessed by DEXA.

Name / AbbreviationDescription

Experimental Factor: Drug Administered

 2,4-Dinitrophenol
2,4-Dinitrophenol (DNP) is a precursor to other chemicals and is biochemically active, inhibiting adenosine triphosphate (ATP) production in cells with mitochondria.
 BAM15
BAM15: N5,N6-bis(2-fluorophenyl)-[1,2,5]oxadiazolo[3,4-b]pyrazine-5,6-diamine, a novel mitochondrial protonophore uncoupler capable of protecting mammals from acute renal ischemic-reperfusion injury and cold-induced microtubule damage
 Niclosamide
Niclosamide: inhibits glucose uptake and oxidative phosphorylation
